NK cells can be used in consolidation therapy for multiple myeloma
To date, multiple myeloma remains an incurable disease and autologous haematopoietic stem cell transplantation is one of the mainstays of treatment, often followed by long-term maintenance therapy with immunomodulators or proteasome inhibitors. However, due to the average age of patients receiving a diagnosis of 65-70 years, the use of autologous HSCT therapy to achieve long-term control of disease progression is not realistic. In addition, there are many serious side effects associated with the long-term use of maintenance therapy, including the risk of haematopenia, thrombosis, secondary malignancy and neuropathy. A promising approach is the use of alternative cell therapy as consolidation after autologous HSCT, which does not have the side effects of maintenance therapy.

Vigilance! Senescent Immune Cells Age Bones
The human skeleton undergoes a continuous remodelling process throughout adulthood to repair small cracks that keep appearing. This remodelling process is precisely balanced between osteoblastic bone formation and osteoclastic bone resorption. However, with age, this homeostasis is markedly compromised in terms of reduced bone remodelling and accumulation of bone marrow fat, which in turn can lead to bone fragility, particularly as bone marrow mesenchymal stem cells (BMSCs), the sole cellular source of osteoblasts, are prone to fat formation rather than bone formation during ageing. Current research suggests that this is caused by mediators in the bone marrow microenvironment, whose primary function is to support and regulate the skeletal function of various cell types, including haematopoietic and mesenchymal cells, in order to maintain homeostasis in the body.

SUMOylation is a reversible post-translational modification associated with cancer. Many post-translational modifications occur on proteins, mainly controlling protein function, localisation and stability, and regulation of proteins by reversible post-translational modifications, such as phosphorylation and ubiquitination, enhances cellular response to stress. Numerous proteomics studies have revealed differences in post-translational modifications between healthy and tumour tissues: phosphorylation and ubiquitination are abnormally elevated in a wide range of cancers, and many protein kinase inhibitors as well as inhibitors of the ubiquitination pathway have been clinically used for the treatment of different types of tumours; however, their specificity is still a major obstacle to cancer therapy.
